David Walliams and Matt Lucas will be reuniting for a radio version of the comedy, 12 years after the show last aired on TV.
Titled Little Brexit, the one-off episode will be broadcast on BBC Radio 4 on Halloween.
The comedy will feature much loved characters including "the only gay in the village" Dafydd, ill-educated teen Vicky Pollard and best friends Lou and Andy.
Original cast members including Gavin and Stacey's Ruth Jones and Buffy The Vampire Slayer's Anthony Head will also make cameo appearances.
Former Doctor Who Tom Baker will narrate.
The original character-based sketch show gently mocked different classes of British society, taking its inspiration from the phrase 'little Englander'.
Starting out on the radio in 2000, it ran for two years before getting its own TV show.
A hit with audiences, it ran for three series and several specials, and was watched by more than nine-and-a-half million viewers at its peak.
An American version of the show, called Little Britain USA, also ran for one series on HBO in 2007.
Commissioning editor for Radio 4 comedy, Sioned Wiliam, said: "Matt Lucas and David Walliams are uniquely talented writers and performers as well as one of the UK's most loved comedy duos.
"It's a privilege to have them back on Radio 4, where Little Britain first began almost 20 years ago."
